The traditional soft agar colony formation assay monitors anchorage-independent cell growth by measuring proliferation in a semisolid culture media in tissue culture dishes after 3-4 weeks, followed by manual counting of colonies. This method is cumbersome and not amenable to high throughput, and the manual counting of colonies is subjective because with varying colony sizes, it’s difficult to determine meaningful results. The CytoSelect™ 96-well Cell Transformation Assay (Soft Agar Colony Formation) eliminates subjective manual counting of colonies with a much shorter assay time. Cells are incubated only 6-8 days in a semisolid agar media before being solubilized, lysed and detected by the provided CyQuant GR Dye in a fluorescence plate reader, making it a more quantitative and higher-throughput method. The shorter incubation time makes it possible to assay cells transiently transfected with oncogenes or siRNA. Each kit is sufficient for five 96-well plates.
